Have been able to draw by hand for a bit and I wanted to get back into vector illustration (which I love just as much). For vector work, I have different processes depending on what I´m doing. Sometimes I do a rough sketch by hand, scan it and retrace/color it using my computer. Other times that I´m working with silhouettes I take pictures and trace the outlines of the object/animal/person. Others I just start drawing freely with the mouse. Here what I did, was collect a bunch of different inspiring images from the internet (color, shape & style related), placed them all around my illustrator work area, and started freely drawing.
FYI I own no drawing tablet and therefore do all my vector art with a mouse. I have, however, used them at past jobs and will probably get one in the future. I haven´t had a real need for one yet, nor do I think they are completely necessary if one already has enough skills with a mouse.
A little side note: I was watching the bonus features of one of the Mad Men seasons I own a while ago and there was this section about the character illustrations being used in all sorts of official stuff for the show. Dyna Moe, the illustrator of these wonderful cartoons, uses a mouse!
